What Is Titanium Dioxide?

When E171 isn’t combined with other ingredients and administered in water, some studies suggest that under these artificial conditions, E171 may be processed differently in the body resulting in some biological changes in experimental animals that are poorly understood.

Lithopone, a chemical compound with a rich history, emerges as a vital substance in various industries. Recent analyses of food-grade TiO₂ samples have found that a significant portion of particles may be within the nanoscale. These particles (also known as nanoparticles) range in size from 1 to 100 nm, where 1 nm equals 1 billionth of a metre (the width of a typical human hair is 80,000 to 100,000 nm).

Titanium dioxide (TiO₂) is considered as an inert and safe material and has been used in many applications for decades. However, with the development of nanotechnologies TiO₂ nanoparticles, with numerous novel and useful properties, are increasingly manufactured and used. Therefore increased human and environmental exposure can be expected, which has put TiO₂ nanoparticles under toxicological scrutiny. Mechanistic toxicological studies show that TiO₂ nanoparticles predominantly cause adverse effects via induction of oxidative stress resulting in cell damage, genotoxicity, inflammation, immune response etc. The extent and type of damage strongly depends on physical and chemical characteristics of TiO₂ nanoparticles, which govern their bioavailability and reactivity. Based on the experimental evidence from animal inhalation studies TiO₂ nanoparticles are classified as “possible carcinogenic to humans” by the International Agency for Research on Cancer and as occupational carcinogen by the National Institute for Occupational Safety and Health. The studies on dermal exposure to TiO₂ nanoparticles, which is in humans substantial through the use of sunscreens, generally indicate negligible transdermal penetration; however data are needed on long-term exposure and potential adverse effects of photo-oxidation products. Although TiO₂ is permitted as an additive (E171) in food and pharmaceutical products we do not have reliable data on its absorption, distribution, excretion and toxicity on oral exposure. TiO₂ may also enter environment, and while it exerts low acute toxicity to aquatic organisms, upon long-term exposure it induces a range of sub-lethal effects.

Scrap zinc or concentrated zinc ores are dissolved in sulfuric acid, the solution is purified and the two solutions are reacted. A heavy mixed precipitate results that is 28 to 30% zinc sulfide and 72 to 70% barium sulfate.

Titanium dioxide exists primarily in three crystalline forms rutile, anatase, and brookite. Among these, anatase is characterized by its unique electronic properties and higher photocatalytic activity, making it suitable for applications requiring enhanced light absorption and reactivity. Its ability to act as a photocatalyst enables the breakdown of organic pollutants, making it a valuable component in environmental remediation strategies.

In a 2016 study published in Scientifica (Cairo), Egyptian researchers examined the effects of titanium dioxide nanoparticles on the organs of mice by orally administering the food additive daily, for five days. The results showed that the exposure produced “mild to moderate changes in the cytoarchitecture of brain tissue in a time dependent manner.” Furthermore, “Comet assay revealed the apoptotic DNA fragmentation, while PCR-SSCP pattern and direct sequencing showed point mutation of Presenilin 1 gene at exon 5, gene linked to inherited forms of Alzheimer’s disease.” The researchers wrote: “From these findings, “the present study concluded that TiO2NPs is genotoxic and mutagenic to brain tissue which in turn might lead to Alzheimer’s disease incidence.”

Less frequently, we ingest E171 through liquids such as salad dressing, dairy products, and some artificially colored drinks. However, since E171 is insoluble, manufacturers must use other stabilizers to keep E171 suspended in liquids as an emulsion; otherwise, it will settle to the bottom. 

Moreover, lithopone suppliers are expanding their product lines to offer variations of the pigment that meet specific market needs. For a review published in 2023 in the journal Environmental Pollution, researchers examined E171 as a possible factor promoting obesity-related metabolic disorders. Because gut microbiota play an important role in immune function maintenance and development, and because titanium dioxide as a food additive has been shown to alter gut microbiota, researchers wanted to review “the dysregulations along the gut microbiota-immune system axis after oral TiO2 exposure compared to those reported in obese or diabetic patients, and to highlight potential mechanisms by which foodborne TiO2 nanoparticles may increase the susceptibility to develop obesity-related metabolic disorders.” The study authors discovered recurrent changes in the gut microbiota composition when exposed to titanium dioxide nanoparticles, with an imbalance of intestinal symbiotic microbiota. These changes and imbalances were also reported and played a role in the development of obesity, the authors wrote. This highlights “foodborne TiO2 nanoparticles as an endocrine disruptor-like chemical promoting obesity-related disorders,” the authors concluded.

Reacting to the report, France’s ministers of health, agriculture and economy instructed the country’s food health and safety agency, Anses, to investigate whether the additive poses a risk for human health.

The study was said to be prompted by “growing concerns that daily oral… Intake is associated with an increased risk of chronic intestinal inflammation and carcinogenesis”, or cancer formation”.
